The Resistance gene is a member of the NBS-LRR (Nucleotide Binding Site - Leucine Rich Repeat) class and codes for a protein that recognizes specific proteins of Phytophthora infestans termed elicitor proteins which are the causing agent of late-blight disease. Rpi-amr3 is a coiled-coil nucleotide-binding leucine-rich repeat (CC-NC-NLR) resistance protein with 887 aa from Solanum americanum.
Upon binding this effector, Rpi-amr3 undergoes ATP-dependent conformational change via its functional domain, Nucleotide-binding and Apaf-1, R-protein, and CED-4 (NB-ARC) and activates helper Nucleotide-binding and Leucine-rich repeat receptors (NLRs) (e.g., NRC2/4). This results in the formation of resistosomes that trigger defense signaling. Recognition of the elicitor proteins will lead to local plant cell death and will thus prevent further development of the pathogen Phytophthora infestans.

Unlike many R genes that are specific to P. infestans, Rpi-amr3 exhibits broad-spectrum resistance across multiple Phytophthora species such as P. infestans, parasitica, P. cactorum, P. palmivora, P. megakarya, P. litchii, P. sojae, P. lateralis, and P. pluvialis, expanding its utility beyond late blight resistance.
